Principles of the Test
|
This type of assay is called “sandwich” assay because the analyte to be measured is bound between the coating (capture) antibodies and the detection antibodies. The sandwich format is sensitive and robust. Here, monoclonalAngiotensin converting enzyme 2 antibodies, showing high specificity toward a single epitope, are used as capture antibody to provide specificity and allow fine detection and quantification of antigens; biotin-conjugated monoclonal Angiotensin converting enzyme 2antibodies, recognizing epitope(s) that are not recognized by capture antibodies, are used as detection antibodies. When the antigen binds to the capture antibody, the epitope recognized by the detection antibody must not be obscured or altered. Capture and detection antibodies that do not interfere with one another and can bind simultaneously are considered a matched pair and are suitable for developing a sandwich ELISA. The biotin-conjugated detection antibodies are further reacted with streptavidin-conjugated HRP-linked enzyme, which can develop a color upon the addition of TMB substrate.
