Generanl Information

Gentamicin (Gentamicin, GM) is a kind of multi-component antibiotic produced by a

Micromonospora strain, and is mainly used for the serious infections such as sepsis,

respiratory infections, pneumonia, intestinal infections, biliary tract infections, urinary tract

infections, soft tissue infections. Toxicological effect of gentamicin is like other

aminoglycosides, with strong renal toxicity, liver toxicity, and even ototoxicity with big dose

of parenteral use. Gentamicin have cross-resistance phenomenon with neomycin,

kanamycin, and is considered to have the greater antibiotic toxicity

Principles of the GM ELISA

The assay operates on the basis of indirect competitive ELISA. The microwell plate is

coated with gentamicin complete antigen. The gentamicin standard or the sample solution

was added, then the specific anti-gentamicin antibody is added. The gentamicin protein

conjugates coated on the microplate and gentamicin in standard or sample solution

competes with the antibody. The second antibody conjugated with horseradish peroxidase

(HRP) is used as a tracer. Tetramethylbenzidine (TMB) was added as a chromogen

substrate. The TMB reaction is stopped by the addition of stopping solution, which converts

the solution to a yellow-coloured product. The colour intensity is measured at 450nm

wavelength. The concentration of gentamicin in the sample is inversely proportional to light

intensity.

Kit Contents

· 1 microtitre plate (96 wells, coated with gentamicin antigen).

· 6 bottles of Gentamicin standard solution (0.25ng/ml, 0.5ng/ml, 1ng/ml, 5ng/ml,

10ng/ml, 20ng/ml) (0.5ml / vial).

· 1vial of Gentamicin antibody (5 ml). Ready to use.

· 1vial of HRP secondary antibody (10ml). Ready to use.

· 1 bottle of color buffer A(6ml)

· 1 bottle of color buffer B (6ml)

· 1 bottle of stopping solution (5ml)

· 1 bottle of sample dilution buffer (20ml)

· 1 bottle of wash buffer (25ml, need to be diluted 10 times before use)
